If you're coming from Mumbai take the first train out at 5:40 AM  which starts from CST station and reaches Karjat at 8:00 AM. Take the local 8-seater rickshaws (Tum-Tum) to Peth Village and explore the fort till lunchtime. You can have lunch at the local hotels around the fort or carry your food. Post lunch, take the Tum-Tum to Kondana Village and alight near the caves, you can explore the caves till evening and leave by 5:00 PM before it gets dark. Catch a return train from Karjat station around 6:00 PM, and you'll reach Mumbai by 9:00 PM. In case you're going for trekking, choose Kondana fort and spend the day there. You can stay for the night in the hotels or camp out and visit the Peth Fort, next day in the morning.
